Abstrak

Robusta coffee is one of the coffee varieties that holds strategic value in empowering the people's economy. To significantly increase coffee yield, it is necessary to propagate the plants in order to ensure the availability of high-quality Robusta coffee seedlings from superior clones to support high productivity. Plant propagation through tissue culture is one alternative to mass-produce coffee plants. The use of cytokinin PGR (Plant Growth Regulator) has an effect on the appearance of globular structures, the number of globular embryo stages, the weight of embryo groups and callus, and the percentage of globular embryo development—one of the cytokinins used being kinetin. The aim of this study is to determine the effect of kinetin PGR on the development of somatic embryos in Robusta coffee. This research used a Completely Randomized Design (CRD) with a non-factorial (single-factor) approach. The study was conducted in the Plant Tissue Culture Laboratory of Jember State Polytechnic from November 2023 to March 2024. The planting material used was embryogenic callus. The study involved 4 treatments: 0 ppm kinetin, 2 ppm kinetin, 4 ppm kinetin, and 6 ppm kinetin. Based on the results, the 4 ppm kinetin formulation was found to be the most effective among the treatments, as it accelerated the formation of globular embryos and increased the final callus weight. However, the use of kinetin was still not optimal for the torpedo and cotyledon formation stages, as kinetin primarily supports early stages such as initiation and callus formation.
