Optimalisasi Beberapa Metode Sterilisasi Pada Eksplan Biji Sorgum (Sorghum Bicolor L.) Secara In Vitro

Sorghum is a cereal crop with the potential for utilizing all parts of the plant. In Indonesia, sorghum cultivation is still overshadowed by other commodities with higher economic value. Another factor influencing productivity in sorghum cultivation is the limited availability of superior seeds. One technique that can support these development efforts is tissue culture to produce new superior plants. One crucial aspect for the success of tissue culture activities is the explant sterilization process. Selecting the appropriate explant sterilization method involves choosing the right type of disinfectant and the duration of immersion. This research aims to analyze the appropriate sterilization method and its effect on the in vitro growth viability of sorghum seed explants to produce superior seed varieties. This research used a Completely Randomized Design (CRD) Non-Factorial with 6 treatment levels: S1: 70% Alcohol + Copper Oxide 2 g, S2: 70% Alcohol + Cloro 25% + Copper Oxide 2 g, S3: 70% Alcohol + Clorox 25% + NaOCl 3% + Copper Oxide 2 g, S4: 70% Alcohol + Copper Oxide 2 g, S5: 70% Alcohol + Cloro 25% + Copper Oxide 2 g: 70% Alcohol + Clorox 25% + NaOCl 3% + Copper Oxide 2 g. The research results showed that the combination of 70% Alcohol + Clorox 25% + NaOCl 3% + Copper Oxide 2 g effectively reduced contamination. The use of Clorox and NaOCl helped suppress the contamination rate with a contamination percentage of 6.67%.
