Abstrak

Cattleya orchids are among the most economically valuable ornamental plants; however, their in vitro propagation process is costly, primarily due to the use of commercial media such as Murashige and Skoog (MS), which is relatively expensive. Therefore, it is necessary to explore alternative, low-cost, and readily available materials that can reduce production costs without compromising the effectiveness of plantlet growth. This study aimed to evaluate the effect of mung bean (Vigna radiata) sprout extract at various concentrations on the growth of subcultured Cattleya plantlets and to determine the most effective concentration. The research was conducted at the Biotechnology Laboratory of Universitas Mercu Buana Yogyakarta using a Completely Randomized Design (CRD) with five treatments and five replications. The treatments included: MS medium (control), MS + extract at 50 g L⁻¹, 100 g L⁻¹, 150 g L⁻¹, and 100% mung bean sprout extract. Observed parameters included the number of leaves, number of shoots, time of shoot emergence, and the number of explants forming shoots. The results showed that mung bean sprout extract influenced plantlet growth. The 50 g L⁻¹ concentration produced growth comparable to MS medium, while the 100% extract medium yielded the highest fresh weight, indicating its potential as an economical alternative in orchid tissue culture.
